**Background**
Sphingosine kinases (SphKs) are critical enzymes that regulate the balance of sphingolipids, which play pivotal roles in cell proliferation, survival, and apoptosis. Among its isoforms, sphingosine kinase 2 (SphK2) has been implicated in various pathological processes, including the progression of several malignancies. Dysregulation of the sphingosine-1-phosphate (S1P) pathway often contributes to tumor growth and resistance to therapy, making SphK2 an attractive therapeutic target for cancer intervention. By selectively inhibiting SphK2, it is possible to modulate intracellular S1P levels and induce apoptosis in malignant cells. In this context, we will introduce a selective SphK2 inhibitor – K145.
**Definition**
K145 hydrochloride is a selective, substrate-competitive, and orally active SphK2 inhibitor with an IC50 of 4.3 μM and a Ki of 6.4 μM. According to the K145 description, this compound is inactive against SphK1 and other protein kinases, ensuring high specificity for its target.
**In Vitro and In Vivo Studies**
The K145 biological activity has been extensively evaluated in various experimental models. In terms of K145 in vitro studies, treatment with K145 (0-10 μM) for 24-72 hours significantly inhibited the growth of U937 cells in a concentration-dependent manner. Specifically, a concentration of 10 μM for 24 hours was found to significantly induce apoptosis in U937 cells. Furthermore, K145 (4-8 μM) treatment for 3 hours resulted in a decrease in the phosphorylation of ERK and Akt. It was also observed that treatment with 10 μM K145 caused a decrease in total cellular S1P levels without significantly affecting ceramide levels.
Regarding K145 In Vivo efficacy, the compound demonstrated potent antitumor activity in animal models. In BALB/c-nu mice injected with U937 cells, daily oral gavage of K145 (50 mg/kg) for 15 days significantly inhibited the growth of U937 tumors. Notably, no apparent toxicity was observed at this dosage. These results highlight the potential of K145 as a therapeutic agent for K145 Cancer research. In conclusion, K145 is a selective and orally active SphK2 inhibitor that induces apoptosis and exhibits potent antitumor activity.
